Abstract
Objective To observe the changes of sarcoplasmic reticulum Ca 2+ adenosine triphosphatase (SR·Ca 2+ ATPase) gene(SERCA2a) expression in the myocytes under the different conditions of thyroid function. To reveal the effects of Triiodothyronine (T 3) on the gene expression and its regulation in myocardial protection. Methods The rats were divided into normal thyroid function group (group A) and hypothyroidism group (group B), each group were subdivided into normal group, control reperfusion group and T 3 reperfusion group. Using an isolated rat heart working model, the relative mRNA levels of SERCA2a in different groups were determined by Northern Blot analysis. Results The mRNA levels declined markedly in the hypothyroidism groups, in the control reperfusion group and in T 3 reperfusion group. However, the levels increased significantly in the T 3 reperfusion group as compared with the control reperfusion group. Group A:(0.73±0.008) vs (0.62±0.008),group B:(0.27±0 003) vs (0.21±0.003),( P 0.01),the gene expression level correlated closely with myocardial function. Conclusion SERCA2a gene expression depression may be an important mechanism in myocardial ischemic reperfusion injury. T 3 could enhance the expression. Through up regulating gene expression, it should be helpful to protect myocytes from ischemic reperfusion injury, and improve myocardial function. To reveal the mechanism of myocardial ischemic reperfusion injury in the molecular level, and to take useful measures to regulate gene expression may be feasible.
