RAPD Analysis on Genetic Structure of Atractylodes lancea

Abstract
OBJECTIVE To study the genetic structure of Atractylodes lancea and discuss its effect on the genetic polymorphism of geoherbs,Mao-cangzhu.METHODS 47 individuals from 3 populations,which were divided into 5 nongeoherb sub-populations and 2 geoherbs sub-populations,of A.lancea were analyzed by RAPD with 20 random primers.The genetic structure of A.lancea was study according to the polymorphism,here and genetic mutation.RESULTS ①77 bands were polymorphic among the total of 94, pps was 81.91%,I was 0.413 2 and he was 0.274 3 in A.lancea.②All of the 11 individuals of goherbs,which included sub-population 6 and 7,clustered together.③Nested analysis showed variations within populations,between sub-populations and between populations of A.lancea were 76.74%,11.58% and 11.68%,respectively.CONCLUSION There are high genetic diversity in A.lancea.Then the major of the genetic variation are distributed in populations,all though there are obviously genetic variation between populations.Finally,genetic distances among the individuals of Mao-cangzhu,which are the geoherbs of A.lancea,were small.Mao-cangzhu can cluster together both in individual and population level.Genetic factors play an important role in forming Mao-cangzhu geoherbs and there are genetic variations between Mao-cangzhu and the others of A.lancea during the long term adaptation to the environment.
High genetic diversity in Atractylodes lancea, with most variation distributed within populations, and genetic distances among individuals of Mao-cangzhu geoherbs are small, indicating genetic factors play an important role in their formation and long-term adaptation to the environment.
