[Objective] The research aimed to explore cryopreservation of Scortum Barcoo sperm.[Method] The optimum concentrations of NaCl and KCl in dilution were firstly determined according to the sperm activity.The sperm cyopreservation trail was carried out using DMSO and Glycerol as cryoprotectant,and then the resurrection ratio and activity of sperm were determined.[Result] The optimum concentrations of NaCl and KCl in dilution were 0.750% and 0.025% respectively.The optimum cryopreservation result was that the resurrection ratio and activity were 68.2% and 60.7% respectively under the condition of 10% DMSO and 5% Glucose in cryoprotectant.[Conclusion] The concentrations of NaCl and KCl in dilution have a obvious effect on sperm motility.The cryopreservation result of DMSO cryoprotectant is better than that of Glycerol.The preservation effect could be improved by adding appropriate amount of glucose in cryoprotectant.
