Anti-fertility Effect of Peppermint Oil on Male Mice

Abstract
Objective To investigate the anti-fertility effect of peppermint oil on male mice. Method 60 adult healthy male KM mice were randomly divided into three experiment groups and one control group. The mice of three experiment groups were orally administrated with peppermint oil mixed with 0. 5% sodium carboxymethyl cellulose at the concentrations of 0. 135 g /( kg·d),0. 27 g /( kg·d) and 0. 54 g /( kg·d),respectively,and the mice in control group were orally administrated with 0. 5% sodium carboxymethyl cellulose. After one week,adult female KM mice were lumped with experimental male mice at the ratio of 2∶1,and the male mice were treated by peppermint oil till day 19. Finally,the blood of male mice was collected to determine the biochemical indices. The male and female mice were anatomized and the rate of pregnancy was observed. The organ coefficient of testis and epididymis,sperm survival rate and deformation rate,pathology and histology of the testis and epididymis tissues were studied. Results Compared with the control group,the pregnancy rate of peppermint oil treated male mice was decreased; the organ coefficient of testis and sperm survival rate was declined,and the sperm deformation rate was significantly increased. Under the optical microscope,the testis and epididymis of peppermint oil treated male mice were damaged. The spermatogenic cells of testis were loosely arranged and the interstitial was reduced. The seminiferous tubules were infiltrated by macrophages. Although normal structure of epididymal duct was observed,there was hardly any sperm in the lumen. Conclusions The anti-fertility effect of peppermint oil can be used to inhibit the spermatogenesis and destroy the spermatogenic tissue of male mice.
Peppermint oil can inhibit spermatogenesis and destroy spermatogenic tissue in male mice, reducing pregnancy rates and sperm survival rates.
