Abstract
The synthesis of 5-amino-N-(2-hydroxyethyl)-2,4,6-triiodoisophthalamic acid,which is used as the intermediate for iodinated X-ray contrast agents,is described in this paper.5-Amino-N-(2-hydroxyethyl)-2,4,6-triiodoisophthalamic acid was synthesized from monomethyl 5-nitroisophthalate by a three-step procedure,i.e.:amidation,reduction,and iodization.The total yield was 79%.The reduction reaction condition was studied.The optimal reaction conditions were conducted at a ratio of m(Pd)/m(5-nitro-N-(2-hydroxyethyl)-isophthalamic acid)=0.001,a reaction temperature of 75 ℃and a reaction time of 6 h using water as the solvent.The conversion of 5-nitro-N-(2-hydroxyethyl)-isophthalamic acid was 100%.
