Abstract
2,4-Dinitrophenylhydrazine is suggested as a quantitative colorimetric reagent for the determination of microgram quantities of prednisone. The sensitivity of this reagent is compared with that of other steroid colorimetric reagents such as blue tetrazolium and isonicotinic acid hydrazide. Extraction procedures are presented which isolate prednisone from formulations containing salicylamide, acetaminophen, aspirin, and ascorbic acid, where the ratio of analgesics to steroid is 300 to 1. 2,4-Dinitrophenyl-hydrazine quantitatively determines prednisone in concentrations as low as 1.26 × 10−6M.
