Abstract
In a citric acid-Na_2HPO_4 buffer solution of pH 2.2, tetrabromofluorescein (TBFS) combines with BSA to give a compound having its absorption maxima at 530nm which is 16nm longer than the absorption maxima of TBFS itself. Based on this new and simple reaction system, TBFS is used as a spectrometric probe in the molecular absorption spectrometric determination of protein. Beer′s law is obeyed in the range of 0.11~60.0μg of BSA per ml of solution. Precision test at the concentration level of 15.0μg·ml~(-1) of BSA, the RSD′s (n=10) found is 1.26%. The Sandell′s sensitivity of this reaction is 0.094μg·cm~(-2). In its application to the direct determination of protein in a control blood serum sample, a results of 35.4±2.4mg is obtained which agrees very well with the standard value (36.9mg) of this control sample. Recoveries found were in the range of 97.0%~108.3%.
