Abstract
Synthesis of 4,4'-diaminodiphenylsulfone (dapsone, DDS, DADPS [Fig. 1]) by Fromm and Wittmann^ in 1908 was the basic step for the worldwide therapeutic use of this compound in leprosy starting in 1941 (Faget et al.^). in addition, dapsone was introduced into chemoprophylaxis of malaria especially as a potential curative agent against chloroquine-resistent strains of plasmodium infections.'' Furthermore, dapsone can be assumed to be an effective therapeutic agent in cutaneous leishmaniasis.'' Actually, dapsone is used in combination with trimethoprim for therapy of Pneumocystis carinii pneumonia in patients with acquired immunodeficiency syndrome (AIDS).^'^ Since 1950, sulfones have also proved to be effective in the treatment of noninfectious inflammatory diseases of the skin such as dermatitis herpetiformis, pustulosis subcornealis, acropustulosis infantilis, and some other dermatoses like necrotizing angiitis that are mainly characterized by neutrophil amplification or circulating immune complexes or both.'''^^ Most of these dermatoses are thought to be of autoimmune nature or demonstrate a prominent infiltrate of polymorphonuclear leukocytes (PMNs) or both. The observed dapsone-induced inhibition of both PMN-mediated cytotoxicity and mitogen-stimulated lymphocyte transformation has provided valuable clues to our understanding of the action mode of dapsone as well as new insights into the pathogenesis of certain inflammatory disorders.
