Abstract
Objective To investigate the effect of a neuropeptide Y1 receptor agonist,[Leu31,Pro34]NPY,on cardiomyocyte hypertrophy in rats.Methods Cultured neonatal SD rat cardiomyocytes were stimulated with [Leu31,Pro34] NPY in various concentrations.3H-Leu incorporation was used to assess protein synthesis rate.Real time quantitative PCR was used to detect ANF and β-MHC mRNA expression in cardiomyocytes.Results [Leu31,Pro34]NPY (10-9 mol/L,10-8 mol/L,10-7 mol/L)caused a significant increase in protein synthesis rate in dose-dependant manner,compared with the control cells(P 0.001).β-MHC mRNA expression was promoted remarkably by [Leu31,Pro34]NPY 10-7 mol/L(P0.001),but ANF mRNA expression was not changed significantly.Conclusions In cultured neonatal cardiomyocytes,[Leu31,Pro34]NPY increases the protein synthesis rate and induces the expression of β-MHC.These data suggest that NPY stimulates cardiomyocyte hypertrophy via Y1 receptor.
