Abstract
Objective:To evaluate the efficacy and tolerahility of topical tacrolimus compared with hydrocortisone and vehi- cle tor atopic dermatitis.Methods:Electronic searches of the Cochrane Library Medline,OVID and CBM.Randomized con- trolled trials(RCTs)of topical tacrolimus reporting efficacy outcomes or tolerability were included.Results:Thirteen random- ized controlled trials were included.Tacrolimus was more effective than vehicle in 12 weeks,0.1% taerolimus was more effec- tive than 1% hydrocortisone acetate and more effective than combined treatment with 0.1% hydroeortisone butyrate(used on trunk) plus 1% hydrocortisone acetate(used on face)in 6 months study.Besides,0.1% tacrolimus was as effective as 0.1% hydrocortisone butyrate in three weeks study.In addition,0.03% tacrolimus was more effective than 1% hydrocortisone acetate but less effective than 0.1% hydrocortisone butvrate.No serious adverse reactions were reported except irritating and burning sensation of skin.Conclusions:Topical tacrolimus is more effective as compared with placebo treatments for atopic dermati- tis;0.1% topical tacrolimus may be more effective than potent topical corticosteroids and may have a place for the patients with atopic dermatitis in a 6-month-period of treatment.However,in the absence of long-term studies on topical tacrolimus, the more researches ahout its safety should be done for a longer period.
