Abstract
(+)-7-(trans-3-Aminomethyl-4-trifluoromethyl-l-pyrrolidinyl)-1-cyclopropyl-6-fluoro-l,4-dihydro-8-methoxy-4-oxoquinoline-3-carboxylic acid hydrochloride (S34109) (I in Fig. l) is a novel quinolone antibacterial agent, having a potent activity against gram-positive bacteria, including methicillin-resistant Staphyrococcus aureus (MRSA).1'2 In order to determine the absolute configuration of (I), an X-ray analysis was performed on an (R)-a-methylbenzylamine salt (II in Fig. 1) of the synthetic intermediate, (-)-traps-l-benzyl-4-trifluoromethylpyrrolidine-3-carboxylic acid (III in Fig. 1). The single crystal of (II) was prepared as follows. To a solution of the racemate of (III) in hot methanol, (R)-amethylbenzylamine was added, and the resulting mixture was left standing at room temperature overnight. The precipitate was collected by filtration, and an optically pure diastereomeric salt (II) was readily obtained by
