Abstract
The structure of 3-deoxy-D-manno-octulosonic acid was confirmed by its reduction to 3-deoxy-D-glycero-D-galacto-octonic acid and 3-deoxy-D-glycero-D-talo-octonic acid which were identified by comparison with authentic acids. 3-Deoxy-D-manno-octulosonic acid was synthesized in good yield by the oxidation of 3-deoxy-D-glycero-D-galacto-octonic acid and 3-deoxy-D-glycero-D-talo-octonic acid prepared by the action of sodium cyanide on 2-deoxy-D-manno-heptose.A method has been developed for the quantitative determination of 3-deoxy-D-manno-octulosonic acid, involving gas chromatographic analysis of the direct products of trimethylsilylation using bis-(trimethylsilyl)trifluoroacetamide for its conversion to volatile stable derivatives. The gas chromatographic method has been used for the identification of 3-deoxy-D-manno-octulosonic acid in several gram-negative bacterial lipopolysaccharide preparations.
