Abstract
Methyl 4-(5-formyl-2-methoxyphenoxy)benzoate was one of the most important intermediate for the total synthesis of bisbibenzyls,which is a series of natural products with versatile biological activities.In this work,methyl 4-(5-formyl-2-methoxyphenoxy)benzoate was synthesized from methyl 4-bromobenzoate and iso-vanilline by condensation reaction,and the reaction conditions affecting the yield,such as catalyst,condensing agents,mixed condensing agents and reaction time had been screened.It was found that cupric oxide and the mixture of calcium carbonate with potassium carbonate was the best catalyst and condensing agent,respectively.The best material inputting ratio was proved as follows:n(iso-vanilline):n(methyl 4-bromobenzoate):n(calcium carbonate):n(potassium carbonate):n(cupric oxide)=1:1.05:0.2:0.8:0.3,The reaction time is about 16 hours.
The best catalyst and condensing agent for methyl 4-(5-formyl-2-methoxyphenoxy)benzoate synthesis are cupric oxide and a mixture of calcium carbonate and potassium carbonate, with a reaction time of 16 hours.
