Abstract
Marked hypocalcemia followed ip and sc (460 mg/kg) or oral (920 mg/kg) administration of 2-thiophenecar boxy lie acid to rats. The drug also caused a fall in serum phosphorus. The maximum decline in serum calcium occurred at 2 hr after drug administration. It was still at this low level (7.6 mg/100 ml) at 4 hr and had returned to the control level by 8 hr. The hypocalcemic effect of 2-thiophenecarboxylic acid was retained after removal of the parathyroid glands, both the parathyroid and thyroid glands, the kidneys or the pancreas, indicating that the presence of these organs is not necessary for its action. In parathyroidectomized rats, 2-thiophenecarboxylic acid antagonized completely the hypercalcemic effects of parathyroid hormone and vitamin D2. No overt toxicity to the drug was seen. The effects of 2-thiophenecarboxylic acid on mineral metabolism are similar in many ways to those produced by thyrocalcitonin.
