Abstract
Formamidines are of continuing preparative interest, 1 in part because of their demonstrated pharmacological activities (Figure 1). The latter include antimicrobial, 2,3 pesticidal, 4 – 6 mono-amine oxidase inhibitory, 7,8 a 2 -adrenergic agonistic, and prostaglandin E2 inhibitory 9,10 prop-erties. Formamidines are significant compounds for their applications in histidine catabolism, 6 purine biosynthesis 6 and as precursors to N -heterocyclic carbenes. 11 In coordin-ation chemistry, formamidines and their anions act as ligands to coordinate with transition metals via monodentate chelation or bridging modes. 12 – 16 In organic synthesis, formamidines can be used as support linkers in solid-phase synthesis, as protecting groups for primary amines, auxiliaries in asymmetric synthesis and in the preparation of dicarboxyalkylthiacarbo-cyanines. 17 – 21 They serve as reactive synthons for a number of chemical transformations 22 and in polymer synthesis as amidine-containing building blocks. 23
