Abstract
Fluorescein-isothiocyanate is a new type of Edman reagent 1,2) in which fluorescein chromophore is at tached to isothiocyanate group instead of phenyl group of phenyl-isothiocyanate (PITC). The strong fluorescence of the reagent enabled the Edman's peptide sequence analysis far more sensitive than ordinary PTC method. Fluorescein-thiohydantoin (FTH-)amino acids have been identified by thin-layer chromatography on silica gel3) or micropolyamide.r) The separation was satisfactory except FTH-Leu and -Ile . Here we report high speed liquid chromatography (HSLC) of FTH-amino acids. HSLC method gives quantitative information which enables us to dis tinguish the signals of the terminal amino acid and contaminated amino acids which increase by increas ing steps of successive degradation of peptide.
