Abstract
3-(diethoxyphosphoryloxy)-1,2,3-benzotriazin-4(3H)-one (DEPBT) is an organophosphorus compound developed by our group which can be used as an efficient coupling reagent for peptide synthesis [1]. Recent studies have demonstrated that DEPBT can be used in either solution-phase or solid-phase synthesis [2]. The advantages of DEPBT are that it has good solubility and causes less than 1% racemization. N-protected amino alcohols and N-protected peptide alcohols have attracted attention, as important synthetic intermediates. Amino alcohols can be used as chiral ligands to catalyze asymmetric catalytic reactions. In this paper DEPBT is used for the first time in the synthesis of peptide alcohols. Our experimental results demonstrated that DEPBT can be used not only in peptide synthesis, but also in the synthesis of amide between amino acid and amino alcohol. DEPBT selected the couplings of amino groups in amino alcohols and the hydroxy groups need not be protected.
