Abstract
Qualitatively, there was no difference in the metabolism of the three substances. No indication of a specific metabolic reaction with the carcinogenic compound was found. The dimethylamines were demethylated and the resulting amines acetylated. 4′-hydroxy-, N-hydroxy-, 3-hydroxy-acetylamines and 3-hydroxy-amines were identified as hydroxylation products. 3-or 4′-hydroxy-dimethylamines, 4′-hydroxy-amines as well as acetylation products of methylamines were not detected. In urine the 3-hydroxy-amines and 4′-hydroxy-acetylamines were excreted as sulfates, the N-hydroxy-and 3-hydroxyacetylamines as glucuronides. 80–90% of the fecal metabolites were unconjugated. The results indicate N-hydroxylation of methylamines and amines beside the acetylamines. Quantitatively, not only the amounts of metabolites produced, but also the excretion patterns were different. The three acetylamines were N-hydroxylated to a comparable extent, assuming ortho-hydroxy-acetylamines to be rearrangement products of N-hydroxy-acetylamines or one of their derivatives. 5 hrs after trans-DAS administration metabolites are bound to precipitable material in the liver to an extent (25 nMole/g liver) comparable to known liver carcinogens. This indicates an activating metabolism in a tissue not susceptible to tumor induction in our experiments. The results indicate the very differentiated chemico-biological interactions involved after administration of the three structurally closely related amines, and how difficult it will be to find reliable correlations between these interactions and the biological effect. More precise knowledge about the pharmacokinetics of these substances will be necessary. Qualitativ ist der Stoffwechsel der 3 Substanzen gleich. Hinweise auf eine für die carcinogene Verbindung spezifische Stoffwechselreaktion ergaben sich nicht. Die verabfolgten Dimethylamine werden stufenweise zu den Methylaminen und Aminen entmethyliert und die primären Amine acetyliert. Als Hydroxylierungsprodukte wurden die 4′-Hydroxy-, N-Hydroxy-und 3-Hydroxy-acetylamine sowie die 3-Hydroxy-amine identifiziert. Bei den untersuchten Verbindungen werden keine 3-und 4′-Hydroxy-dimethylamine und keine 4′-Hydroxyamine gefunden. Die 3-Hydroxy-amine und 4′-Hydroxy-acetylamine werden im Harn als Sulfate, die N-Hydroxy-und 3-Hydroxy-acetylamine als Glucuronide ausgeschieden. Die Faeces enthalten praktisch nur unkonjugierte Metaboliten. Aus den Befunden wird geschlossen, daß nicht nur die Acetylamine, sondern auch die Methylamine und Amine N-hydroxyliert werden. Quantitativ bestehen erhebliche Unterschiede im Stoffwechsel der 3 Vergleichsverbindungen, nicht nur was den Anteil bestimmter Stoffwechselwege, sondern auch was die Ausscheidungsmuster betrifft. Unter der Annahme, daß die ortho-Hydroxy-acetylamine durch Umlagerung aus N-Hydroxy-acetylaminen oder Derivaten davon entstehen, wird eine vergleichbare N-Hydroxylierung der 3 Acetylamine gefolgert. 5 Std nach Applikation waren trans-DAS Metaboliten im gleichen Umfang an fällbares Material in der Leber gebunden (25 nMol/g Leber) wie bekannte Hepatocarcinogene. Das deutet auf einen Aktivierungsstoffwechsel in einem Gewebe, in dem keine Tumoren entstehen. Die Untersuchungen machen deutlich, wie differenziert die chemisch-biologischen Wechselwirkungen bei 3 Verbindungen so ähnlicher chemischer Struktur zu betrachten sind, und wie schwierig es ist, eine gültige Korrelation zwischen diesen Wechselwirkungen und dem biologischen Effekt zu finden. Eine noch viel subtilere Kenntnis der Pharmakokinetik der interessierenden Substanzen wird dazu nötig sein.
