Abstract
Abstract Dimethylsulfoxide (DMSO) dissolves tetramethylrhodamine isothiocyanate (TRITC) as well as fluorescein isothiocyanate (FITC) rapidly and completely. Furthermore, gamma globulin is unaffected by dilutions of up to 30% DMSO in carbonate buffer. These two facts facilitate the labelling of antibodies with fluorochromes with special reference to TRITC, which cannot be completely dissolved in other non-denaturing agents. Labelling with crystalline TRITC produced readily characterizable but poorly performing conjugates. On the other hand, amorphous TRITC resulted in high quality conjugates, which could not be properly characterized. The quality of FITC conjugates was equal to that of conventionally produced conjugates, the only advantage being the rapid dissolution of FITC in DMSO.
