Abstract
OBJECTIVE To study the effect of dl-tetrahydropalmatine (dl-THP) on various rat gastric ulcers and analyze its possible mechanism.METHODS The gastric mucosal damage was induced by pyloric ligation, restraint, indomethacin and acetic acid in rats, dl-THP 15, 30 and 45 mg·kg-1 was given ig bid for 5 doses. Then the number of gastric ulcers or the gastric ulcer area, the secretion of gastric juice, hydrochloric acid and pepsin activity were determined respectively. The stomach blood flow in normal and (dl-THP treated rats was also examined.RESULTS dl-THP 15,30,45 mg·kg-1 protected the gastric mucosa against the damages with a dose-dependent manner. In chronic gastric ulcer model, the inhibitory rates of gastric ulcer of dl-THP and ranitidine 30 mg·kg-1 were 75.8 % and 67.7 % , respectively. However, the secretions of gastric juice, HC1, and pepsin by gastric mucosa were not influenced by dl-THP. Moreover, dl-THP increased stomach blood flow significantly.CONCLUSION dl-THP protected various experimental gastric ulcers by increasing stomach blood flow and maybe partly by antagonizing gastric DA receptors.
