Abstract
Objective:To observe the effect of compound naphthoquine on the ultrastructure of erythrocytic stage of plasmodlum berghei ANKA strain and compare with naphthoquine phosphate, dihydroartemisinin and trimethoprim.Methods:NIH mice were infected with plasmodium berghei ANKA strain and administered ig with compound naphthoquine, dihydroartemisinin, trimethoprim and naphthoquine phosphate respectively. Blood samples were collected at 1st,2nd,4th,8th,12th and 24th hour after drug administration, and the ultra-thin sections were mounted, stanined and examined under electron microscope. Results:The effect of compound naphthoquine on the ultrastructure of erythrocytic stage of P. berghei ANKA strain demonstrated that there is a potent and rapid lethal action on the late trophozoites of erythrocytic forms by interfering the physiological function of membranous phase system. After 2h medication, most of late trophozoites showed progressive morphological changes such as swelling, breaking and vacuolated, the interspace appeared with fusion of food vacuoles, the mitochondrial membranes swollen, separated and stripped and the malaria pigments deformed. After 4h exposure to compound naphthoquine,the degenerated parasites increased. After 12h medication,the structure of most of late trophozoites was destroyed.Conclusion:The effect of compound naphthoquine on the ultrastructure of erythrocytic stage of plasmodium berghei ANKA strain is more prompt and stronger than other relevant drugs.
