Abstract
Objective To study the effect of herbal medicine dracorhodin perchlorate on collagen synthesis in scar-derived fibroblasts and evaluate new drug for human hypertrophic scars. Methods The fibroblasts were cultured as an experimental model and the effect of dracorhodin perchlorate were detected. The cell proliferation was determined by MTT assay, and the collagen production by photometric determination. The mRNA expression of typeⅠand Ⅲ procollagen was detected by RT-PCR. Results It was shown that exposure to dracorhodin perchlorate could increase the inhibiting rate of fibroblasts by MTT assay and dependent on concentration and time(P0.05), otherwise significantly reduce the soluble collagen content in the culture medium(P0.05). The results of RT-PCR showed the expression of typeⅠand Ⅲ procollagen was decreased along with the increasing of the concentration of the drug(P0.05). Conclusion dracorhodin perchlorate could significantly inhibit the proliferation and the collagen synthesis of fibroblasts in vitro.
