Abstract
The antithyroid activity of orally administered analogues of goitrin (D,L-5-vinyl-2- oxazolidinethione) and related compounds was assessed in the rat using the indices of an increase in thyroid weight and a reduction of radioactive iodine uptake. Selected compounds were also tested for effects on radioactive iodine uptake after parenteral administration. Propylthiouracil was found to be 150 times as potent as goitrin after oral administration but only 40 times as potent after parenteral administration. The only other compound found to be effective by mouth without obvious toxic effects was 4-ethyl-2-oxazolidinethione. Its potency was similar to that of goitrin. (Endocrinology 81: 88, 1967)
