Abstract
Objective To study the effects of dimethylamiloride(DMA) on heart function in normal rat isolated hearts.Methods With Langendorff-perfusion,the changes of the parameters reflecting heart function in normal rat isolated hearts were observed to estimate the effects of DMA.Results ①In normal rat isolated hearts,DMA(2.5~20 μmol/L) improved the heart function.The extent of enhancement of 2 μmol/L DMA on LVSP-LVDP,+dp/dt_(max) and-dp/dt_(max) was 8.17%、5.22%、10.15%,of 5 μmol/L DMA it was 7.59%,8.84% and 10.93%,of 10 μmol/L DMA was 9.27%,10.53% and 11.48%,and of 20 μmol/L DMA it was 8.37%,11.19% and 13.31%,respectively.②The effects of DMA on heart function started to present at min 2,maximized at min 6,then declined gradually,and disappeared entirely at min 10.Conclusion DMA(2.5~20 μmol/L) may improve normal rat isolated heart function with certain perfusion time.
