Abstract
Objective: To study the effect of a pyrazol chemical compound,pyr3,the selective blocker of transient receptor potential canonical 3(TRPC3) on pressure overloaded rats for their left ventricular hypertrophy(LVH)in vivo.Methods: The pressure overloaded rats model was established by abdominal aortic constriction.30 male SD rats With weight of 200~240g were randomly divided into 5 groups: ① Sham operation group,② Operation control group,③ Sham operation with whole-course medication group,④ Operation with later medication group and ⑤ Operation with wholecourse medication group.n=6 in each group and all rats were treated for 6 weeks.The left ventricular mass index(LVMI) and left ventricular myocardial transaction diameter(LVMTD) were calculated;the mRNA and protein expressions of TRPC3 and calcineurin Aβ were examined by RT-PCR,Western blot analysis and immunohistochemistry respectively.Results: Compared with Sham operation group,Operation control group presented the increased LVMI,LVMTD,higher mRNA and protein expressions of TRPC3 and calcineurin Aβ.In Sham operation with whole-course medication group,the left ventricular hypertrophy related indexes were no real changes while the mRNA and protein expressions of TRPC3 and calcineurin Aβ were obviously increased,all P0.05.Compared with Operation control group,Operation with whole-course medication group showed decreased hypertrophy related indexes and decreased mRNA and protein expressions of TRPC3 and calcineurin Aβ,all P0.05.In Operation with later medication group,the hypertrophy related indexes and mRNA and protein expressions of TRPC3 and calcineurin Aβ were decreased,allP0.05,while TRPC3 mRNA and protein expression was similar.Conclusion: pyr3 treatment may prevent pressure overloaded rats for their LVH at certain point,and decrease the upregulation of TRPC3 and calcineurin Aβ.
