Abstract
Effects of methylenedianiline were investigated in the rat, the rabbit, and the dog.In the castrate female rat, this compound produced adrenal hypertrophy, enlargement of the uterus, and was goitrogenic. In the dog, methylcnedianiline effected a marked inhibition of adrenal 17-hydroxycorticoid secretion. In the immature female rabbit, this compound elicited progcstational proliferation of the endometrium which was shown to be adrenal-dependent. Adrenalectomy as well as cortisone-treatment blocked the progestational effect of methylenedianiline. Inasmuch as these effects are also characteristic of amphenone, these studies provide evidence that the methyl ketone structure is not essential for the responses observed. However, methylenedianiline lacks the marked anesthetic effect of amphenone.
