Abstract
OBJECTIVE To study the effects of Panax quinquefolium 20s-protopanaxdiolsaponins (PQDS)extracted from leaves of Panax quinquefolium on experimental ventricular remodeling in rats. METHODS The ventricular remodeling model was induced by myocardial infarction for 4 weeks in rats with left anterior descending coronary occulusion. After 4 weeks of treatment with PQDS, hemodynamic changes, biochemical analysis and morphological parameters were determined in ventricular remodeling rats with myocardial infarction. RESULTS After being treated by PQDS (in a dosage of 25~100 mg·kg~ -1 ip for 4 weeks),the maximum left ventricular pressure rising and dropping rates (±dp/dt_ max )and their left ventricular systolic pressure corrected values(±dp/dt_ max /LVSP)were increased significantly. The left ventricular end diastolic pressure (LVEDP), the left ventricular volume(LVV), the left ventricular long and short axis(LVLA and LVSA) , the left ventricular absolute and relative weight (LVAW and LVRW) were reduced significantly, but there were no significant influences on the right ventricular absolute and relative weight (RVAW and RVRW) , the heart rate (HR), the left ventricular systolic pressure(LVSP), mean artery pressure (MAP) and body weight (BW). In addition, the serum lipid peroxidation (LPO) content, myocardial angiotensin Ⅱ(AngⅡ) and epinephrine (E) contents were declined, and the serum superoxide dismutase(SOD), catalase(CAT) and glutathione peroxidase (GSH-Px) activities were increased markedly.CONCLUSION PQDS can efficiently prevent and treat ventricular remodeling in rats with myocardial infarction, which may be related to the inhibition of promoting growth action of AngⅡ and CA,and the elimination of the free radicals in rats with ventricular remodeling induced by myocardial infarction.
