Abstract
Objective To investigate the effects of glaucocalyxin B on cell proliferation and cell cycle of retinoblastoma(Rb) cell line in vitro.Methods MTT method was used to detect the effect of glaucocalyxin B with different doses(2.5 μmol·L-1,5.0 μmol·L-1,10.0 μmol·L-1,20.0 μmol·L-1 and 40.0 μmol·L-1) on growth of human Rb cells at different time points(24 hours,48 hours).The effects of glaucocalyxin B on the cell cycle and apoptosis of Rb cells were observed by propidium iodide(PI) staining and flow cytometer,respectively.Results Glaucocalyxin B could inhibit cell proliferation in each dose group.The optical density went down with and the apoptotic rates went up with increasing of dose.The IC50 value for the inhibition of Rb cells by glaucocalyxin B for 24 hours and 48 hours were 79.15 μmol·L-1 and 10.88 μmol·L-1,the dose-effect relationship was obvious.The apoptotic rates of Rb cells after treated with glaucocalyxin B with different doses(2.5 μmol·L-1,5.0 μmol·L-1,10.0 μmol·L-1,20.0 μmol·L-1 and 40.0 μmol·L-1) for 24 hours were(6.21±0.91)%,(10.21±2.06)%,(16.83±2.61)%,(25.17±2.11)% and(35.29±1.04)%,respectively,and(2.01±0.03)% in control group,which for 48 hours were(7.94±3.01)%,(12.08±2.43)%,(20.11±2.37)%,(35.18±3.48)% and(51.20±2.93)%,respectively,and(2.41±0.73)% in control group.The apoptotic rate of Rb cells was obviously increased after treated with glaucocalyxin B with a certain dose dependent manner.Conclusion Glaucocalyxin B can inhibit the proliferation of Rb by inducing the apoptosis of Rb cell strains.
