Abstract
Introduction The new thioxanthene derivative, thiothixene (Iqavane; P-4657B), has been reported to be an unusually effective antipsychotic drug in chronic schizophrenic patients (SUGEl~M~N et al., 1965; S~Pso~ and IQBAL, 1965; GALLANT et al., 1966; BISHOr et al., 1966; WOLPEaT et al., 1966; SIMEON et al., 1967). The present report describes several psychopharmacological actions of thiothixene and compares them with those of chlorpromazine, prochlorperazine, and the phenothiazine analogous to thiothixene, thioproperazine. Geometric isomerism resulting from the unsaturated 9-position of thiothixene made possible the resolution of isomers and an investigation to determine whether the psyehopharmaeological activity of thiothixene, the isomer with the side chain cis, relative to the nuclear substituent, is shared by the trans isomer (P-4657A). Pronounced and consistent differences in pharmacological activity between geometric isomers of other 9-unsaturated thioxanthenes have previously been reported (MCLLE~ NIELSEN et al., 1962).
