Abstract
Durch Einwirkung von Brom auf L-Phenylalanin wird unter Erhaltung der Konfiguration am α-C-Atom ein Gemisch von Mono- und Dibromderivaten dieser Aminosaure gewonnen, das sich in Methanol-Wasser an Sephadex LH-20 vollstandig auftrennen last. Je nach Bromierungszeit erhalt man (Tabelle 1) bis zu 29% L-2-Bromphenylalanin (1), 36% L-4-Bromphenylalanin (2), 2% L-2,3-Dibromphenylalanin (3), 13% L-2,5-Dibromphenylalanin (4) und 5% L-3,4-Dibromphenylalanin (5). Die Zuordnung der Isomeren geschah durch ihre NMR-Spektren (Tabelle 2). Zur weiteren Charakterisierung wurden die Elutionsvolumina in der Aminosaureanalyse (Abbildung 2), die optischen Drehwerte, die IR- und UV-Spektren (Tabelle 3, Abbildung 3) bestimmt. A Simple Preparation of Isomeric Bromo-L-phenylalanines A mixture of monobromo- and dibromo-L-phenylalanines, obtained by bromination of L-phenylalanine, is completely resolved on LH-20 Sephadex in a methanol water mixture. Depending on reaction time the yield is up to 29% for L-2-bromophenylalanine (1) and 36% for L-4-bromophenylalanine (2). The isomers L-2,3-dibromophenylalanine (3), L-2,5-dibromophenylalanine (4) and L-3,4-dibromophenylalanine (5) are obtained in yields of up to 2%, 13% and 5%, respectively (Table 1). The substances were identified by their NMR spectra (Table 2), and further characterised by amino acid analysis (Fig. 2), UV spectra (Fig. 3), IR data and optical rotation (Table 2). All compounds were obtained optically pure.
