Abstract
The problem is to balance the known disadvantages of the hepatotoxicity of the estolate against the stated advantage of better absorption, bearing in mind the reasonable indications for oral erythromycin therapy. On the many occasions this matter has been examined by the Committee, consideration has been given both to data retrieved from the literature and submitted by the companies concerned. Since erythromycin was developed in the early 1950s and was not subjected to the detailed pharmacological and clinical investigation now required of new drugs, much of what has been accepted as fact is actually only surmise or inference. The purpose of this communication is to explain the current status of the problem and the difficulties which have been encountered.
