Abstract
Abstract In order to obtain reproducible and characteristic pyrolysis of thermally labile samples such as proteins without causing any undesirable denaturation before the final thermal degradation, a vertical microfurnace-type pyrolyser was used. The high-resolution pyrograms of the samples were obtained with a fused-silica capillary column. The general pyrolysis behaviour was first studied with various aliphatic and aromatic α-amino acids, then the high-resolution pyrograms of proteins were interpreted in terms of the compositional analysis of amino acid residues in the samples. The simultaneous determination of aromatic amino acid residues such as Phe, Tyr and Trp in various enzymes was successfully carried out by use of 300 μg of the samples. Aliphatic amino acid residues such as Leu and Ile in various enzymes were also determined.
