Abstract
Abstract Three thymine analogues, 5-acetyluracil, 5-(1-hydroxyethyl)uracil, and 5-vinyluracil have been added in turn to a thymine-deficient medium in which a thyminedependent strain of Escherichia coli has been grown. Only 5-vinyluracil has been incorporated into the DNA and its presence was confirmed by enzymatically hydrolysing the DNA to nucleosides for chromatographic comparison with authentic samples. The relative rates of deoxynucleoside formation from the bases by the enzyme, pyrimidine deoxyriboside phosphorylase confirmed that 5-vinyluracil was the only one of the three analogues likely to compete with thymine for incorporation into the DNA.
