Abstract
Objective To investigate the influence of clausenamide on blood sugar,glycosylated hemoglobin and hippocampal glial fibrillary acidic protein(GFAP) mRNA expression in diabetic rats.Methods The diabetic rats were randomly divided into model group,insulin treatment group(10 U.kg-1 insulin,hypodermic injection),high dose clausenamide group(50 mg.kg-1 clausenamide,intragastric administration) and low dose clausenamide group(25 mg.kg-1 clausenamide,intragastric administration),fifteen rats in each group.Fifteen rats in control group were treated with citrate buffer solution by intraperitoneal injection.The diabetic rats model produced by injection streptozotocin were treated by clausenamide.After 3 months,blood sugar and glycosylated hemoglobin were observed,RT-PCR was used to detect mRNA expression of GFAP in hippocampus of diabetic rats.Results Compared with control group,the levels of blood sugar and glycosylated hemoglobin inmodel group increased obviously (P0.01),but there was no statistical difference in the levels of blood sugar and glycosylated hemoglobin between the model group and clausenamide group(P0.05).The results of RT-PCR showed that the expression ofGFAP mRNA in hippocampus of model group was higher than that of control group(P0.01).The expressions of GFAP mRNA of clausenamide groups and insulin treatment group were lower than that of model group(P0.01).Conclusion Clausenamide can significantly inhibit GFAP mRNA expression in hippocampus,which may account for the protective effect.But clausenamide can not degrade the levels of blood sugar and glycosylated hemoglobin.
