Abstract
Abstract Ethyl α,γ-difluoroacetoacetate, ethyl fluoroacetate, and diethyl fluorooxalacetate inhibited citrate formation from oxalacetate by rabbit kidney cortex mitochondria preparations. The ethyl difluoroacetoacetate inhibition was not competitive with oxalacetate. Accompanying this inhibition, there was an accumulation of oxalacetate and pyruvate. The in vitro action of ethyl fluoroacetate, contrary to its action in vivo , is due to the lack of esterase activity in the mitochondria preparations used. Evidence is presented which indicates that sodium monofluoroacetate has an inhibitory action at the “acetate” stage, prior to the condensation reaction, as well as its inhibitory action due to the formation of fluorocitrate.
