Abstract
Objective To study the lipid-lowering effect of Asperosaponin VI,a triterpene saponin isolated from Dipsacus asper Wall in ob /ob mice with Non-alcohol fatty liver disease( NAFLD). Methods To induce NAFLD,a high-fat diet was given to ob /ob mice. Asperosaponin VI( 30,60,120 mg /kg) were intraperitoneal administered daily for 4 weed. The effects of Asperosaponin VI on serum levels of total cholesterol( TC),triglyceride( TG),low-density lipoprotein cholesterol( LDL-C),high-density lipoprotein Cholesterol( HDLC) were detected. Meanwhile,the body weight of mice need to be examed,and the liver tissue were stained with Hematoxylin-eosin( HE) and were histologically analyzed. In the present study,we use oleic acid induced HepG2 cells developed a cell model of steatosis,and the effect of Asperosaponin VI on lipid droplet formation was examined with Bodipy 493 /503 staining method. Results In ob /ob mice models with NAFLD,60 and 120 mg /kg of Asperosaponin VI significantly decreased TC,TG and LDL-C level. Therefore,the levels of HDL-C was significantly increased. Asperosaponin VI remarkably decreased body weight and liver index. HE staining showed that the fat particles in liver cytoplasm were remarkably reduced by Asperosaponin VI. medium and high dose group. Furthermore,Asperosaponin VI high dose group reduced the lipid droplet formation in HepG2 cells. Conclusion Asperosaponin VI has antilipemic activity and relieves fatty degeneration in liver cells,suggest that it may be a potential agent for treatment of NAFLD
