Abstract
Terbinafine is a member of the allylamine class of antim\ cotics, which are established as inhibitors of fungal ergosterol biosynthesis.' As with the related compound naftifine/ a large body of experimental evidence indicates that specific inhibition ofthe enzyme squalene epoxidase (Fig. 1) is the primary mechanism of action of tcrhinafine.' The present paper reviews this evidence with the aim of elucidating the biochemical basis for the antimycotic properties of terbinafine, with particular emphasis on the following three aspects:
