Abstract
Abstract Polyanetholesulfonate (PAS) inhibited Factor Xa initiated clotting of bovine plasma. PAS also accelerated thrombin inhibition by antithrombin III. When lysine residues on antithrombin III were modified with 0-methyl isourea, 50% of the heparin co-factor activity was lost. The PAS effect on thrombin inhibition by the modified antithrombin was reduced by the same amount. PAS did not stimulate antithrombin III inhibition of Factor Xa. When PAS was added to a Factor Xa-antithrombin III reaction mixture, the rate of Factor Xa inhibition was reduced. These results indicate that polyanetholsulfonate anticoagulates plasma by stimulating the inhibition of thrombin by antithrombin III while having no effect of the inhibition of Factor Xa by antithrombin III.
