Abstract
Metyrapone is an inhibitor of the adrenal enzyme 11-β-hydroxylase, which results in the blockade of cortisol synthesis. Its uses are primarily for experimental protocols in which the effects of endogenous cortisol are removed for brief periods of time. It can be useful for the diagnosis of Cushing's disease, determining the maximal adrenocorticotropic hormone (ACTH) secretory capacity of the pituitary corticotrophs, or combining with challenges to the hypothalamic-pituitary-adrenal axis to evaluate the importance of cortisol negative feedback.
