Abstract
Abstract High performance liquid chromatography with 9-fluorenylmethyl chloroformate (FMOC) permits detection of nitrogen-containing solutes including ectoine (1,4,5,6-tetrahydro-2-methyl-5-pyrimidinecarbocylic acid) from halophilic eubacteria. To enable ectoine derivatization with FMOC reagent the heterocycle of ectoine was cleaved by mild hydrolysis. The HLPC method described for the detection of compatible solutes is high;y sensitive and allows analysis of single bacterial colonies from agar plates. Consequently this procedure is suitable for a rapid screening for nitrogen-containing solutes in halophilic and halotolerant microorganisms.
