Abstract
Abstract The nitrosation of 12 drugs which contain amine, urea, or simple amide groups have been studied. Nine drugs yielded no detectable N -nitroso derivatives, but niridazole and tolbutamide gave the expected N -nitrosoureas and hydrochlorothiazide gave the N -nitrosamine. The formation of N -nitroso-hydrochlorothiazide showed a fairly high second-order rate constant of 0.76 m −1 sec −1 at pH 1 and 25°C. For tobutamide, nitrosation succeeded only with dinitrogen tetroxide in methylene chloride and the pure product was not obtained reproducibly. Data are reported on the solubility, stability, and acute LD50 in rats of the N -nitroso compounds.
