Abstract
Objective: To study the pharmacokinetics of flucloxacillin sodium capsules after a single oral dose in Chinese Han and Tibetan healthy volunteers.Methods:Ten Han and ten Tibetan healthy volunteers were involved in the study.They were evenly composed of male and female.Each subject received a single oral dose of 500 mg flucloxacillin sodium capsules.The plasma concentration of flucloxacillin was determined by HPLC method.The pharmacokinetic parameters were calculated by DAS 2.1 software and were statistically analyzed by SPSS software.Results:The main pharmacokinetic parameters of flucloxacillin in Han and Tibetan volunteers were as follows: cmax(16.85±7.13) and(14.23±6.04) μg/ml,t max(1.02±0.07) and(0.95±0.04) h,t1/2(1.61±0.24) and(1.45±0.29) h,AUC0-10(61.36±15.63) and(63.02±17.74) μg·h·ml-1,AUC0-∞(65.74±16.87) and(64.36±19.54) μg·h·ml-1.These parameters of male and female volunteers were as follows: cmax(16.93±6.16) and(14.18±7.83) μg/ml,t max(0.93±0.04) and(1.05±0.06) h,t1/2(1.58±0.35) and(1.44±0.21) h,AUC0-10(61.85±17.62) and(63.64±14.75) μg·h·ml-1,AUC0-∞(63.52±16.73) and(65.36±20.04) μg·h·ml-1.Conclusion:The plasma concentration-time curve of flucloxacillin meets a two-compartment open model,and its main pharmacokinetic parameters have no significant differences between Han and Tibetan nationality,or male and female volunteers.
