Abstract
SC-5233, or 3-(3-oxo-l7β-hydroxy-4-androsten-17α-yl) propionic acid γ-lactone, blocked the urinary effects of desoxycorticosterone acetate (DCA) in rats by reversing sodium retention and potassium loss. The mechanism of action appeared to involve reversible competition at the receptor sites. The compound also blocked the urinary effects of aldosterone, hydrocortisone, 21-desoxy- 9α-fluorocortisone and 19-nor-DCA. SC-5233 inhibited the development of DCA-hypertension in rats and reduced the blood pressure in metacorticoid hypertensive rats. SC-5233 did not possess cortisone-like, androgenic, anabolic, or estrogenic activity, nor did it block the effects of standard steroids in these tests.
