Abstract
Phenol is a competitive inhibitor of CO 2 hydration and a noncompetitive inhibitor of HCO 3 − dehydration catalyzed by human carbonic anhydrase II (or C). From these results and the pH dependence of the inhibition it is concluded that the enzyme-inhibitor complex is a combination of neutral phenol with the high pH form of the enzyme. Spectroscopic results suggest that phenol is bound in the hydrophobic part of the active site in the vicinity of the metal ion without displacing zinc-bound OH − .
