Abstract
Z-2-(2-aminothiazol-4-yl)-2-methoxyimino acetyl chloride hydrochloride was synthesized from 2-(2-aminothiazol-4-yl)-2-methoxyimino acetic acid via acidification and chlorination. Phosphorus pentachloride was replaced by thionyl chloride and N, N-dimethyl formamide as chlorinating reagent in chlorination. The effect of reaction temperature and solvent on the yield of Z-isomer and E-isomer was investigated. Results show that, by using thionyl chloride and N, N-dimethyl formamide as chlorinating reagent and dichloromethane as solvent and taking -15°C as reaction temperature, Z-iomer crystals with pale white color and stable to temperature can be obtained with an overall yield of 70%. Under the aforementioned condition, no E-isomer solid with yellow color and unstable to temperature is obtained basically. The title compound used as an amino acetylating reagent was further reacted to yield Cefepime hydrochloric acid, the crystals obtained has white pale color and is stable to temperature, and has no E-isomer too. The title compound and Cefepime hydrochloric acid obtained were characterized by 1HNMR. The procedure proposed is simple and mild, which is suitable for industrial production.
