Abstract
ABSTRACT Following the administration of 3H-labelled alanyl-alanyl-prolyl-valine chloromethyl ketone to mice by stomach tube, a portion of the labelled chloromethyl ketone can be recovered in an active form in lung wash. Animals given a single oral dose of 300 μg of the synthetic elastase-inhibitor, 15 min prior to introduction of 6 units of porcine pancreatic elastase into the lungs, were completely protected against alveolar deformation (as judged from mean linear intercept values 4 weeks later). Treatment with the inhibitor at times earlier than 15 min before enzyme challenge or from 15 min to 48 h after enzyme challenge did not protect.
