Abstract
Abstract A rapid and relatively specific assay method for 2,3-diphosphoglyceric acid (DPG) is described. The procedure involves the stoichiometric conversion of 2,3-DPG to glyceraldehyde-3-phosphate with measurement of the resulting change in reduced nicotinamide adenine dinucleotide (NADH) absorbance or fluorescence. 2-Phosphoglycolate is utilized to activate the 2,3-DPG phosphatase activity present in muscle monophosphoglycerate mutase.
