Abstract
Objective:To investigate the interaction between ethyl caffeate and herring sperm DNA (HS-DNA).Methods:The interaction between ethyl caffeate and herring sperm DNA was investigated by cyclic voltammetry,differential pulse voltammetry and ultraviolet absorption spectrometry.Results:It was shown that ethyl caffeate exhibited a pair of electrochemical redox peaks in 0.1 mol·L-1 phosphate buffer (PBS,pH 7.4) at glassy carbon electrode.Meanwhile,both the cathode and anode peak currents decreased with increasing the amount of HS-DNA,and the formal potential shifted negatively.Conclusions:The interaction between ethyl caffeate and HS-DNA was mainly of an electrostatic one in phosphate buffer (pH 7.4),by the determination of electrochemical parameters in the absence and the presence of HS-DNA,it was deduced that a non-electroactive supramolecular compound was formed between ethyl caffeate and HS-DNA.The molar binding ratio and the binding constant of ethyl ferulate into HS-DNA were calculated to be 2 and 9.8×106,respectively.Ultraviolet absorption spectroscopy has validated the above conclusion.
