Abstract
Clopidogrel has become the most widely used antiplatelet drug second to aspirin. Clopidogrel itself is a prodrug that is absorbed in the intestine and partially converted into 2-oxo-clopidogrel by a cytochrome P450 monooxygenase-dependent metabolism. The intermediate metabolite 2-oxoclopidogrel is further hydrolyzed to the active thiol metabolite of clopidogrel that irreversibly binds to the platelet Gi-coupled adenosine diphosphate (ADP) P2Y12 receptor.
