Abstract
Of a series of diarylalkanes tested for depressor activity on renal and metacorticoid (post-DCA) hypertension in rats, two nitriles were equally active. These were 2,3-bis(p-hydroxyphenyl)valeronitrile and the corresponding propionitrile. The valeronitrile also displayed estrogenic activity, growth inhibition, and adrenal hypertrophy in rats; large doses induced severe hemorrhagic responses in dogs. At comparable doses of the propionitrile, these effects were minimal or absent. In general, it was found that relatively few alterations in the molecule were consistent with anti-hypertensive activity.
